How to Promote Cell Adhesion
- 1). Culture two batches of endothelial cells from a human umbilical vein in Dulbecco's modifed Eagle's medium and 10 percent fetal calf serum. Place in a cell incubator at 37 degrees Celsius with a ratio of 5 percent CO2 and 95 percent atmospheric air.
- 2). Add a 1-to-10 dilution of blood serum from humans treated for four weeks with 0.033 Mg per Kg of Norditropin (growth hormone) once daily to one batch of cultured cells. Return to the cell incubator at the same settings for 12 hours.
- 3). Add a 1-to-10 dilution of blood serum from humans not treated with Norditropin to the other batch of cultured cells. Return to the incubator at the same settings for 12 hours.
- 4). Measure the concentration of vascular cellular adhesion molecule-1 (VCAM-1) in the two batches by incubating for one hour with rabbit antigoat-Ig-HRP antigen and reading absorbance at 540 Nm with an ELISA reader. The level of VCAM-1 in the Norditropin serum-treated cells will be significantly higher than that in the cells treated with serum from humans not treated with Norditropin. This indicates increased cell adhesion in the Norditropin serum-treated group.
